Well this is not the first mission John made it for them, the first mission you need to find all the posts around Rhodes town because they are escaped from the jail, after find all the posts you need to go where are they camping and talk to them after that if you keep playing the game for maybe a couple days in real time, you will find them sick and they will ask you for medicine, and John will help them and he will tell them to leave.
