Will u explain how direct coomb's test helps to diagnose HDFN,since the mother develops antibody against baby Rh antigen and it stays as unbound one(free floating antibody) in her blood?

It is done with the cord blood of the fetus and hence, Direct coomb's test will be +ve

I did not give anti d injection at the time of my first delevery. That's why I had 2nd baby abortions, but i didn't give anti-d injections, 3rd baby i gave 28week anti-d injections when I was a 3rd child. After having a baby, I was given anti-d injections within 24 hours. I've got antibiotics now..?

I have a question. I hope it's not a stupid question. You said complements needs cell surface, then in indirect coomb, isn't there RBCs too? I wonder if the fact that the antigen is known in indirect has anything to do with it. I love your videos, btw.

The RBC which is used in indirect combs test are washed and used ( these are pre processed) for the process while in direct coombs they are directly taken from the patient's body . Yes that's why we use the words direct and indirect coombs test. Hope that answers your questions. Sorry if I wasn't able to elucidate it 😊

Sir, if Antibodies attached to Surface, doesnt that mean that Antibodies were circulating? If Antibodies attached to surface are also circulating (which must) then why at indirect Coombs test cant we also test for example autoimmun heamo. anemias? IGg and Igm must be also circulating. Can you eloberate please? Or because If we see even Antibodies in Serum that doesnt necesarrily mean that also attached to Cells so in second stage we must perform direct Coombst test. Is that why doesnt make sense to use indirect Coombst test to study AIH or Drug induc. anemias etc?
