ImageJ tutorial: Band Intensity

    1. Open your image file
    2. Select the widest band in the image as the first band by using “Rectangular Select” tool.
    3. Selected area’s height should be twice of the widest band on the image.
    4. Press “CTRL +1” to mark the first selection and number 1 will appear inside the box selection.
    5. Move the mouse inside the first box selection until it changes to an arrow shape.
    6. Click and drag that selected box area to the next band in the image.
    7. Press “CTRL + 2” to mark the second box selection.
    8. Repeat step 5 to 7 for the next bands in the image.
    9. After all the bands are selected press “CTRL + 3”
    10. And the histogram of each band will appear.
    11. Select the “draw line” button and draw a line across the lower part of each the histogram.
    12. Select the “Magic Wand” tool and click inside the histogram.
    13. The selected area will be outlined in yellow, and Result window will appear.
    14. The Result windows indicate the quantitive intensity value of the each band.

      Sorry for late reply, invert your image (the band should be black or darker than the background)

      still works the same if you just define (with the line tool) the area not under the curve but above it
