4:25 if temperature for annealing is kept 5’C less than Tm wouldn’t the actual denatured strands tend to reanneal ????
How is their reannealing opposed???
For annealing 55℃ is required for 45 seconds, time can be variable ,when length of dna fragment varies

Primer has both 5 prime and 3 prime both ends, so it can bind complementary to 5 prime end of the template strand also. So how pcr overcomes that situation?
No ..3 prime provide free oh group for binding..so primer get bind to the 3 prime only..

Its not like you are counting 2 primer and using them. Its concentration of primer. 2 types of primer in specific concentration
