Things are much simpler these days. There are many unnecessary steps and techniques included in this video that are not currently needed, and the ultramicrotome has advanced drastically.
I'm glad you did! I started doing ultramicrotome in 1966 using a completely manual Cambridge ultramicrotome, with glass knives, cutting phloem tissue. Very difficult! I moved to the Institute of Ophthalmology in 1971 and finally ended up using a Leica ! I retired in 2011,after seeing many changes in TEM and SEM Happy cutting !

Great video! Thanks for uploading. Music made it even more epic ;)
Really good explanation of the steps involved! Thank you!
This will be great for my students! It's so hard to crowd around a microtome, but this video will help with that! :)

what camera did you use to record this??
Just the camera on my phone
@@danielvocelle1414 did you just mount the phone?
@@junglebaby448 yes, built a holder out of wood to hold it on the objective
How small is 70nm just curious
Smaller than the diameter of the coronavirus. Coronavirus is approx. 100-120nm
i dont think that it is nessesary to make marks for trimming becuase anyway you are not cutting on the mark
I hate the soundtrack. Makes it sound like something horrible is going to happen.
Thanks Mary
Block too far out of the holder.
Dreadful score marks on the sections - throw that knife away ! Fit a new one or use a diamond knife
Happy to report knife was thrown away 10 years ago.
