It isn't, amu is an arbitrary unit made with carbons, we don't know what it is in grams. Except we do somehow, we had discovered moles after all which makes those amu into grams..
The explanation was perfect! We got to know that mass of atoms are measured relatively to mass of carbon-12 atom But my question is: How did we measure the mass of a single atom of carbon-12

That measured with carbon itself. We use carbon because it is the common element
