PI is used as a DNA stain for both flow cytometry to evaluate cell viability or DNA content in cell cycle analysis and microscopy to visualize the nucleus and other DNA containing organelles. 1-if you incubate PI for 30 min most cells will take up the stain. just 5 min enough. 2- EtOH needs to be ice-cold before you add.
That's exactly the point. They're showing pi stain for cell cycle. You permeabilize with etoh and stain long with saturated pi BECAUSE you want every cell to stain to measure DNA content. What you said is correct for pi as a viability stain.
For how long will the sample remain viable after its preparation and before introducing it into the flow cytometer?
Once you fix and permeabilse the cells with ethanol (final concentration to be 70%) in -20*C, there is no relevancy of viability in case of cell cycle analysis.
