30-50 mM Imidazole seems really low for an 8X-HIS elution. Are you sure that’s not non specific binding coming out and your protein is still bound?
We realized there was a typo and it was actually ~150-300!
I feel so spoiled having only ever done purification with an AKTA Pure FPLC.
How pure was your protein after only nickel purification? We always also do a SEC run to polish and put it in the right buffer aswell.
Surprisingly pure! We did dialysis since we can't easily do SEC here. See part 2... th-cam.com/video/Uma0LmM5MSg/w-d-xo.html
