I have one doubt when we place the IPG strip on the well areas, are they treated with SDS? if yes, how we treated it on the strips? or if not so positively charged protein don't move at all so what's the alternative

Are the placements of the anode and cathode arbitrary? Or does cathode have to be inserted in the side with the lowest pH and the anode with the highest? Also, how are the proteins loaded?
