  • This video describes, How to calculate the percentage drug release from tablet dissolution studies. In the pharmaceutical industry, drug dissolution testing is routinely used to provide critical in vitro drug release information for both quality control purposes, i.e., to assess batch-to-batch consistency of solid oral dosage forms such as tablets, and drug development, i.e., to predict in vivo drug release profiles. There are three typical situations where dissolution testing plays a vital role: (i) formulation and optimization decisions: during product development, for products where dissolution performance is a critical quality attribute, both the product formulation and the manufacturing process are optimized based on achieving specific dissolution targets. (ii) Equivalence decisions: during generic product development, and also when implementing post-approval process or formulation changes, similarity of in vitro dissolution profiles between the reference product and its generic or modified version are one of the key requirements for regulatory approval decisions. (iii) Product compliance and release decisions: during routine manufacturing, dissolution outcomes are very often one of the criteria used to make product release decisions.Via wiki

    Hi Sir, In min 13:39 you are summing up concentration 5ml (ug/ml) with cumulative 5 mL (ug), is this correct? does not look like it

    Sir if i take 5 ml from simulated gastric fluid and add further 5ml to to my dissolution medium during invitro study.Still it will be necesaary to calculate cummulative.

    Thanks ...Sir plz tell... If my tablet weight is 200mg and dose of my tablet is 25mg ( actual drug in my tablet is 25mg) so for calculation of % drug release by which value I should divide
    Either by total weight of tablet(200mg) or dose of drug in tablet(25mg)??

      Y to do that, if u increase the media then the dissolution power of medium also get increased it will affect the dissolution profile. Think of saturation concept.

    We can replace the 5ml every time when we take out and also we can maintain sink condition.

      Yes but it can cause error. As fresh medium may interfere with dissolving power or medium. Ex. saturated medium can dissolve less solid then fresh.

      This test is in vitro dissolution study . No need to justify sink condition After performing this test we have to correlate this data with in vivo study wich is known as ivivc

      Both procedures can be done , either add medium or minus 5ml at every point.

      ​@@Pharmacy4uu yes , in franz diffusion cell aparatus for drug delivery , we do the same , as you are taking some amount of sample at every time interval , we need to add the same amount of solution so as to maintain the conditions, or else the behaviour or solution left will be totally different , the replacement coefficient is further added to maintain the calculations
