You can avoid using the ball despawn bug with Nian S2 and a less silly approach for the last wave, but I didn't think it was very interesting (Nian S2 holds for 35 while Penance breaks a ball + a standard wave 2 approach) so I went for the funny spawnkill. If you add better VG you can probably clear without VG attacking enemies, but I didn't think that was interesting cause with that DP output the rest of the stage is trivial. What's interesting about DP challenges isn't spamming more vanguards IMO, it's interesting when it forces you to change up tactics and evaluate different approaches by how expensive they are and when they need to be deployed. In this case, the total amount of DP to spawnkill the middle defenders is much less than the block route. But there is no way to get the DP to afford the spawnkill early on in the clear, and I have to put everything off until the end hoping I'll have enough DP. Every deploy I made was done thinking about how they'd contribute to solving the big problem I put off for the end + breaking enough Kristen balls to stop the boss.

The challenge mode didn't really affect me since I could just run the same strategy anyways since Myrtle, Zima, and Bagpipe just printed enough dp even with the cost increase. I am still in disbelief that this guy kept on with defenderknights despite the limitation. Have to respect that imo.
The balls can despawn if they're not interacted with (or they just despawn anyways?) after a large amount of time has past. You can kind of see this with the "infinite" duration skills like Thorns s3 on Annihilation stages. The orange bar ends up ticking down to like halfway or somewhere around there. Seems like Arknights has an internal cap / limit on how long stages should last so things get wonky when you've stalled on stages long enough.
