    the module under the autosampler called Thermostat which used for temp control of Samples in the auto sampler. normally you may set the temp to 4 degree

    It is the sample cooler under the auto sampler to keep the samples at 4C.

    I guess the thermostated column doesn´t really tranfer the heat to the column / solvents, as the column is not thermically coupled to the heated aluminium block..
    There should be clips to secure the column / press it against the aluminium block..
    Furthermore there are heat exchangers which are screwed to the aluminium block with a capilary in them, so the liquid which passes through them get´s heated / cooled to the correct temperature, alternatively there are capilaries embeeded into the aluminium blocks as well (see plastic caps right / left of them).
    Using the front cover could improve the temperature stability, as the column is not subjected to changing air flow / temperatures in the environment (for example AC system).

    do all of the HPLCs gradients specially prominence of SHIMADZU LC-2030C(its a new model actually) one needs to open the purging valve during purging? we are purging without opening the valve. The respective personnel also says no need to open the valve.

      If you sonicate your solvents you don't need to purge.

    it is possible to separate sample components using HPLC and get those components after finishing of analyzing process or HPLC used only for analysis

      There are so called "Fraction collectors", once the detector detects a peak / fraction, they wait for the peak to reach the fraction collector and put each individual peak into a different vial. The software later on shows you which peak was collected into which vial. This can be scaled up and is called "preparative lc" and commonly uses higher flowrates / more sample / solvents..
      On analyitcal scale LC, as you might imagine if you inject only 10nl of sample a fraction will be less than 10nl of real substance you try to extract, disolved in a lot of solvents.
