I have a question please, who cut hydrogen bonds when restriction enzyme make cohesive cut? Coz to separate the 2 strands we have to break hydrogen bond
Once the phosphodiester bonds have been cut, the hydrogen bonds between the two strands of DNA are automatically broken. This is because the hydrogen bonds are only stable when the two strands of DNA are held together by the phosphodiester bonds
The video is very awesome Sir but i have one doubt, If restriction endonucleases can't cut bacterial dna because of presence of methyl group then how we can cut a Plasmid DNA (i.e it is also contain a bacterial DNA)?

That's not gonna happen
That is why we use type 2 restrictions endonuclease
