First filtration followed by sonication since its role is to remove air traps. If you do first sonication again filtration then environmental air entraps the solution and you will get negative peaks due to air bubbles.

I think that the vacuum created by the vacuum pump not only allows suction/filtering, but also catching any air in the solution. But you're right, first filtering and then sonication seems better. Have you noticed that the filter is smaller than the opening of the bottle and some of the solution may have flowed sideways, bypassing the filter?

What is the standard taken here?? And how did u arrive at the formula. Generally if we have a linear equation of standard samples of different conc we can find out the unknown concentration.
