Red blood cells under the microscope, hypo and hypertonic solutions

  • Red blood cells (RBCs) as seen under the microscope in isotonic, hypotonic and hypertonic solutions. A few white blood cells can also be seen with the red blood cells. RBCs contain hemoglobin, a red pigment that binds to oxygen and allows oxygen to travel around the body.
    When RBCs are placed in a hypertonic salt solution, water moves from inside the cell to the outside by osmosis. This causes the cells to shrink. When RBCs are placed in distilled water, a hypotonic solution, water moves from outside of the cell to the inside, causing the cell to swell and rupture. Once the RBC ruptures, it loses its hemoglobin and becomes transparent and nearly invisible. These ruptured cells are called RBC ghosts and are very difficult to see in the video.
    I use two microscopes recorded through a Nikon D3300 DSLR camera.
    Wards trinocular Microscope (not sure the model) - 40X, 100X, 400X, 1000X
    American Optical One - Ten microscope - 150X, 600X, 1500X
    The magnification of each shot is shown on the bottom right hand corner.

    I’ve been trying to do this for some time. I do not have a microscope but I’m not sure of what or how deep the microscope has to be to be able to read dna but I wanted to do that for myself and check my blood and how it reacts to certain things

      There are about 500-800 red blood cells for each white blood cell. The ratio depends on whether you are a male or female and how healthy you are.

      Hi Rodney,
      blood cells are normally floating in blood plasma which has salts, proteins and blood platelets. In this video, I added saline solution so the cells can float around and look natural. Saline is a 0.9% salt solution.

      I use a microscope that costs about $1,000 and a camera that costs $500. So it costs a good amount to get image quality like this.

      Yes, USD. Microscopes vary a lot in magnification, resolution, function and build quality. You would have to shop around to get what you want. Cells differ in size as well but if want to see the smallest cells like bacteria, you should get a microscope that can magnify to 1,000X.     I looked under my blood under a microscope and i saw a ball with a bunch of little spikes on it. Can anyone answer what that was?

      That may have been a crenated or deformed red blood cell. You can see some at 2:09.

    I'm trying to replicate this with my microscope. Do you use only one drop of blood? This saline solution did you purchase it somewhere or did you just make it by combining water and salt?

      Yes, I only used one drop of blood. I made the saline solution. It is a 0.9% sodium chloride (NaCl) solution. You add 0.9 grams of NaCl to 100 ml of distilled water. Use less NaCl for a hypotonic solution and more for s hypertonic solution. For my video I used distilled water and a 10% NaCl solution respectively.

    I have a question, I hope u can answer me: We say that a red blood cell has 0.9% salt concentration. In chemistry, 0.9% salt is 0.9g per 100ml solution but an RBC can't contain 100 ml/doesn't have this volume. So, how will we justify the 0.9 % salt solution for such a little volume of RBC if we apply our logic according to chemistry? Can we say it like this, salt makes 0.9% of the whole fluid present in a RBC.

      Yes, it’s 0.9% salt by weight divided by fluid volume, which is way less than 100 ml.

      Percentage has nothing to do with the amount of a thing as you describe it.
      0.9g in 100ml are 0.9% but every droplet of this Solution is 0.9 % because it is the ratio of salt to water (in this example).
      To make it a little easier, imagine 50%.
      50g in 100ml are 50%.
      If you take 10 ml from it you will have 5g salt in 10ml water so it stays 50% as you can see because it's just the ratio.
      50/100 is the same as 5/10, just put it in your calculater to see both are 0.5 and this means 50%.
      With apples we would have 100 apples and give half of it away(50%). Now we have 50 apples but can still give half of them away. So from 50 apples 50% is 25. Because it's the ratio of apples I give away to my current number of apples: 25/50 = 0.5.
      So yeah there is just a tiny amount of water in your cells but the ratio stays the same because it is also less salt in it.
