Fun Fact: Ed mentions that a lot of Alchemists encode their research to protect it. Ed disguises his research as a travel journal of all the places he’s been to. Meanwhile Mustang disguises his research as a list of all the women he’s dated, part of the reason he has a reputation as a womanizer.
There is also the theory that Al's research is disguised as the list of things he wants to eat.

Real quick, Ed doesn't use a tattoo to transmute. That "symbol" you are thinking of associated with Ed and Al, which Ed wears on his coat and Alphonse on his armor, is the Flamel symbol (in reference to Nicholas Flamel the real world french Alchemist), and in the context of FMA it is the symbol of their teacher's alchemical style (basically it is them representing who taught them alchemy). Edward does not have this symbol, nor any other symbol, tattood on his body.
Edward does not need a circle or tattoo to transmute, he can in a sense create his own circle simply by clapping his hands together. This allows him to create any kind of specific alchemic circle he can think of and use it at any time so long as he can clap his hands together (this is why Scar specifically destroyed Ed's automail arm in order to stop him from transmuting). Ed is capable of doing this because he saw the Truth, who beamed this information into his head when he crossed the gate. Other alchemists, such as Isaac the freezer from episode 1, had a tattoo'd alchemy circle on his hand to allow him to transmute with just that hand. The symbols by themselves are not enough to transmute, a tattoo has to be an alchemic circle in order to be used to transmute.

4:15 Central is, unsurprisingly, in the center of the country. East city is roughly in the center of the Eastern sector of the country. Not a suburb of central. The show doesn't do a great job showing this, though, as Roy is bouncing back and forth between the two cities a fair amount, with no real distinguishing landmarks.
The great thing about FMAB, when they rise a question, your question will be answered. (very soon, unless it affects a whole story)

East City *is not* a part of Central City. It is its own city in the East. This’ll become clearer later as the geography of the country gets explored more.
