How do we know? We compare to known genomic libraries, cDNA libraries etc with known defective and non defective gene codes. PCR procedures using fluorescent markers allows us to identify the position of each nucleotide in the sequence, if you do this a heap of times it is possible to map out the DNA of the replicated strands. You can also use flow through analysis etc (marker flows past a laser or something to detect fluorescence, its nucleotide and position is identified, we can also check and compare using molecular weight etc. The methods are really quite endless)
