Ghetto Spore Dilution Method (from swab)

How to plate spore dilutions for single spore (haploid) isolation. Once they germinate, monokaryotic (hopefully) colonies will be checked for the absence of clamp connections and subcultured. They'll then be clear for use in mating experiments.

Could you not take this solution and make a LC, then peroxide it to sanitize? 🤔 it would kill anything but endospores and the mycileum... If I'm not wrong that would be a pretty cheap and effective way to use a swab without agar while ensuring some amount of success?

That is the idea behind a multispore syringe (MSS). There is no mycelium in this procedure until it is on agar.

Can I cut a swab with scissors then place in a sterilized glass eyedropper bottle? Do I need to ad the soap?

Polysorbate 20. Tween is a brand name. You can just use a tiny, tiny drop of dishsoap too.
