    If you eliminate the filthy spoon, you will eliminate your biggest vector for contamination in that process... Also, if you just flip the box over before you start, and mist a surfactant (dish soap and water) into the box. Then flip it back over and wait for a minute for everything to settle your box will be more efficient by allowing dust and whatever else is present to stick to the walls. By using alcohol your walls are dry and allowing all the stuff to continue floating around inside your box increasing the likelihood of contamination. Just some advice.

      Yeah his techniques are TOTALLY wrong!
      He should've put on a space suit shot himself out into space and done the operation in the vacuum of space, it's the only right way to get proper inoculation!

    How was the accepting grain prepared? I mean, could the accepting grain be ready for LC at this time?

    Hi I’m relatively new to growing mushrooms I was wondering… is there a limit on how many grain to grain transfers you can do? And how would I create a master plate that can last for years? Would I just take a bunch of spore prints or live cultures to agar from my first batch and use that until I’m out then do the same for the next generation?I’ve heard about how your mycelium or mushrooms can age and become sluggish and how there’s a limit to how many times it can divide with out bad mutations or something like that I just want to make sure I’m doing everything correctly so later on down the road I’m not messing up

      If you can keep everything sterile, and the cultures remain contam free, you can keep going. I once used a single jar of spawn to culture more than 250 quart jars over 5 different grain to grain cultures... they were ALL for P mex sclerotia, and after five months I believe it yielded well over 4 kilo of sclerotia... I didn't have a scale big enough to weigh all of it.

    You said in the video that you used 1 tablespoon of donor grain to inoculate 200g of receiver grain, the 200g receiver grain ,is it when on dry state before soaking in water or the final weight after pasteurization/sterilization? one more thing ''nice video''👏👏👏👏

    I'll be getting ready to do this in the next week or 2 so I'll definitely be rewatching this when I can give it more attention. Thanks. I have full quart size jars, of whole oats almost fully colonized. I was planning on saving 1 quart of each genetic line and doing the grain to grain. How many quart jars would you inoculate with 1 quart jar of colonized Spawn?

      I'd use 2 TBSP of donor grain per quart jar of virgin grain. If my rough math is close, I'm thinking you should be able to inoculate ~25 new jars.

    Hi! Thanks for an awesome content. Question? Where you usually store the jars? Or if you use the grow bag? Just store it in a dark place like cabinet like in your video? It’s my first time growing.. Thanks in advance

      Just needs to be 65-80F, a cupboard is where I keep mine!

    What are you using to disinfect??? You mention iso alcohol, but the stuff you are using has a green or blue tint. All of the 70% iso alcohol I've bought and used has always been clear.

    1 bottle grain spawn is how many can use 5bl bag for block?

      I utilized a fruited 2lb (approximately 1000g) block and divided it into four separate 2lb un-fruited grain bags, using about 200g from the fruited block to each of the new grain bags. Though I'm not sure if this scales to larger bag sizes.

    I'm looking at you from a microbiology point of view and everything looks good except you should have disposable rubber gloves and spraying your hands with the alcohol before opening the jars

      Yes, totally agree with that. However, I choose to accept that risk in favor of using less single-use plastics (gloves) and have experienced very little contamination as a result. I'm certain that using gloves would cut down on my contamination rate though.

      Gloves aren't needed much at this stage as fully colonized mycelium has it's own, strong immune system and is usually resistant to contamination. Also, avoiding single use plastics is advisable.

      @@OneEarthMushrooms I'll use gloves because the energy, materials and time in a couple of wasted jars costs (the planet) more than one pair of gloves that can be recycled.
      Personally, I use all natural, biodegradable latex, but I've thrown used nitrile into the autoclave for reuse.
