Gram staining is possibly the single most important differential stain used to distinguish between different types of bacteria based on their chemical and physical properties. In this video, Biology Professor (Twitter: @DrWhitneyHolden) will cover the Gram stain procedure and discuss how it is applied to determine what type of bacterium is causing a given disease. Great for MCAT Biology Review! For more information on the structural differences between Gram-Positive and Gram-Negative Cell Walls, see Biology Professor's video: • Gram Positive and Gram... For more information on another type of differential stain, the Acid-Fast stain, see Biology Professor's video: • Acid-Fast Stain
explained better in 5 minutes than my 2 hour lab lol THANK YOUUUU
RIGHT!
She explains this really clear!
Burnt out teacher who might have been motivated during one point in his/her life. Everyone has seen those. It's always refreshing seeing motivated teachers.
Better explained than all my days at Pathology block posting 👌🏾
Thanks :)
very good
so if I was asked that which is the gram stain should I say crystal violet or saffranin?
Technically, BOTH are required because it is a differential stain...one is used to show Gram-positive bacteria and one is used to show Gram-negative bacteria. If you want to get picky, you might be able to argue that the crystal violet is the main stain and the safranin is just the counter stain.
it was really very helpful, for the first time I completely understood gram staining, thanks a lot 😃😃
Thank you for such a short concise explanation! It really simplified Gram staining for me
MAM
FIRST WE TAKE A SLIDE
THEN WRITE N AND S ON BOTH SIDE
THEN USE NORMAL SALINE AND SOLLEN DUE TO AIR
THEN USE TO HIT
THEN USE CRYSTAL VIOLET THEN USE IODINE AND SAFRINE
Thanks🌹
Awsome explanation very much better than our college faculties
Thanks mam your method of teaching is so good
perfect explanation. easy to follow. thank you
I'm glad it was helpful! I have a lot of other microbiology videos at this playlist: tinyurl.com/BioProfMicro
Check it out and like my videos. :) THANKS
@@BiologyProfessor You explained gram stain very well
We wash slide after each step, right??
Generally yes
ma'am thank you very very much... this was the most helpful vdo for me in 2018... keep making these cool stuff.. coz only we the students know how it feels when we dont understand anything...cramming remains the sole option but it is disastrous in the long term...
2 minutes ago i was getting frustated but i am so happy and calm ☺😏
my microbiology teacher is the worse. I understood this well, way better than in class. I over stained my bacteria because he was no help. thank you
Thank you for the great explanation , helped a lot :)
Your country name
Thank you so much mam...
thankio for this lecture beacause i know completly for gram staining
Thank you so much 💕
thank you professor ur video is informative n ur so sweet!!
Thank you so much... You just summarised my 2 hours lab session in 5 minutes.. That too bet efficiently... Thank you
Nice explanation love from Pakistan
Thanks for watching :)
Good explained
Thanks mam😍😘😘😘

Thank you for this!!!! Awesome job explaining clearly what’s going on
Thank u so much for this explanation
Thanks ! I appreciate you helped me big time before taking my exam tomorrow !!
Good luck on your exam! If you need to review Gram Positive vs Gram Negative bacteria cell walls before your exam, check out my video on that topic: th-cam.com/video/hIj2c8fZWlM/w-d-xo.html
Thank you sooo much for a very clear explanation about gram stain! :)
You're so awesome and gorgeous
Thank you so much this was so simple to understand
This helped thanks! I passed my lab exam! Thanks for sharing your knowledge!
That's great to hear! Glad I could help. :)
Congratulations! :)
Thanks for your help very nice video
Great vid
Thanks!
Your smart
Thank you :)
good explanation, it is of much aid for me. Thanks.
and she's so pretty😍🥰...
Thank you! Best explanation I’ve heard
You are welcome 🤗
the best biology professor on youtube ! thank you
omg thank u for this explanation!!!
Thank you so so much verey helpful
Very well explained. Good work.
If you accidentally used saffrinin instead of crystal violet but then did all the rest of the steps would they all just look pink? Or does the thick cell wall only bind to the crystal violet?
they would all look pink
such a clear explanation thankuuuu
Thnxx mam... Nice explanation
You have nice voice😍😘😘
Thank you, very helpful!!
Watching all your videos on bacteria the night before my lab test, Thank You a bunch 🙏
I did my gram stain wrong 😔💔 oh Jesus
better luck next time... ;)
Thank you so much!
Thank you doctor. You are a real life saviour before my practical examination
I hope you do very well and get a great grade!
Yup. It went smooth 😀
Good job 👏🏻
Ma'am very good
Ma'am what is your professional field
I mean ...which field of biology
Whats the purpose of decolorizing in gram stain
Decolorizing is practically the most important step (obviously, all are necessary) because the decolorizing step is where you get the differences between cell types. After the crystal violet and iodine, ALL cells are purple. The alcohol decolorizing solution removes the purple from Gram-negative cells, but not Gram-positive cells (unless it’s on too long). Then saffranin is used as the counter stain to make the Gram-negative cells visible so you can see them under a microscope. The saffranin would not show up on the Gram-negative cells if they were still purple because you had skipped decolorizing.
Thank you for your presice and perfect explanation.
Thanks! I also have videos on other stains if you are interested, like Acid Fast stain and Endospore stain. I also have a video on the differences between Gram-negative and Gram-positive bacteria, so view it if you get a chance. :)
How you reached the whiteboard top to write the heading...(Gram staining procedure)
May be you just used a stool or chair
The name Safranin really sounds blue-purple because of sapphire blue stone. And the crystal violet just sounds like some pink illicit pills. So, this thing is really messing up our brain.
Thank u so much 😍❤
Thank you mam
thanks you so much !
Thank you so much!!!
Thank you very much ma'am for this beautiful lecture.
what is a mordant
a substance, typically an inorganic oxide, that combines with a dye or stain and thereby fixes it in a material.
Thank you madam
Thank you!
very interesting!!!
nice, simple explanation. don't forget to underline 'Clostridium difficile'
this is what i needed for my flow diagram always that lab assistance will ask us flow diagram before we do the practicals and am sure tomorrow i will be on the good books of heres thaaaaaaaaaank you and maybe i will score some point to my partner besides shez my crush
good 👍
Ur the best!
Wow thanks!
shouldn't cocci(circle shaped) be the violet one, while the bacilli (rod shaped) be the pink one?
No. The cells in my video are generic examples. You are thinking of E. coli as a pink bacillus and S. aureus as a purple coccus because these are common examples. But there are many Gram-negative (pink) cocci like species of Neisseria (diplococcus) and Haemophilus (coccobacillus). And there are many Gram-positive (purple) bacilli like species of Listeria and Clostridium (both rods). In other words, cocci and bacilli can stain either color, depending on the species.
is the channel free
Yes, you can watch all my biology and chemistry videos for free! :)
👍👍
Maam u liked after two month😇😇 but ur video was awesome
Thanks :)
What will happen if you used the decolorizing solution first instead of the primary stain?
If you swap the order, the decolorizing solution won’t remove the crystal violet from the Gram-negative cells, and the saffranin will not be visible. All cells (Gram-pos and Gram-neg) will be purple.
What happens if you forget the mordant or decolorizer for gram positive and gram negative?
It you forget the mordant, the crystal violet dye won’t stick during the decolorization and everything will end up pink from the safranin. If you forget the decolorizer, everything will end up purple because the crystal violet will be retained in both cell types.
Ur are so lovely
no YOU're lovely
😃
👍
Thank you !

Check it out and like my videos. :) THANKS
Wow awesome
Thank you!
Your voice is very down. And your videos are helpful. Please try to raise you tone next time. Thank you
Thank you for the informative explanation. Could you please explain in the last foto why are the bacteria aggregated in certain areas on the slide? was that only random distribution or there is a certain reason for that?
Thanks
+zaid alhajaj It is a random distribution created when the bacteria were smeared across the slide and fixed to it for staining.
👍👍
❤️
thank you so much mam
Wow that was perfect! Thank you so much!
Glad you liked it! Tell your classmates about my channel so they can learn too. :)
Thankz
Such a good break down. One question though, what is the purpose of the counterstain?
To help see the non Gram positive bacteria better.
It is to see the gram NEGATIVE bacteria better. Since they won’t retain the crystal violet following destaining, you need a counter stain to make them visible.
is there an automated gram staining apparatus?
Good question. I’ve never seen one, but that doesn’t mean they don’t exist, perhaps in some really high tech lab somewhere.
god bless you
thank you
stop lying ussssssssssss//////
...?
Cute, Knows way more bacteria scientific name than me, likes to tutor, and her hair is perfect, well.. I'm in love :|
are you still in love
