    Excellent explanation, I really loved you pointing out what colonies you were looking for, labeling, and such. You definitely have the streaking down to a pro level. The small petri dishes are so cute! The incubator was really helpful, and explaining the upside down condensation. Thanks for posting dude!

    How can you be sure the colonies are haploid? I can imagine two spores might be stuck together and create diploid colonies. Do you need to check for clamp connections with a scope?

      You can check for clamp connections but this is very subjective in my opinion (when you look for clamp connections everything looks like one it takes a bit of practice) the best way to verify is to grow it out.

    Hey Gary, thank you for those videos! i want to ask you about using incubator. I've made one years ago from a cooler with a heating cable on bottom covered by sand, and controlled by thermostat, but i found that dry heat lowers humidity and makes the parafilm tear asap. So i have to incubate the dishes inside of airtight tupperwares inside of the incubator to protect them against low humidity, but as heat comes from below it also causes a lot of condensation in some dishes, so i have to put them upside down to decrease the condensation. I wonder if you have the same problems on your incubator and if you have useful tips about using one. I use plastic wrap too but only for fresh poured uninoculated dishes, for inoculated ones i prefer using parafilm. Affordable incubators don't have means to control humidity i guess, so what is your experience using an incubator with petri dishes? thank you

      Andrés Lorenzo Santana being in colorado I am constantly battling low humidity and always use parafilm without any issues just double wrapping seems to do the trick 👍🏻 the sand might contribute to a lower humidity the coils in the incubator I have are exposed and have racks over them for the plates and rarely turn on unless opening and closing the incubator often. Condensation is also a constant battle and can happen for a number of reasons which is why I store the plates upside down - The highest quality incubators that I have used before do not supplement humidity and just maintain temperatures more steadily.     Any chance you could put a single spore one a microscope slide with a well filled with a bit of agar and watch that growth under magnification? And then sealing the slide with a cover slip to prevent contamination

      I would love to do this in the future! The hardest part would be isolating one single spore in a microscopic field - but I'm sure it can be done! MUSHLOVE

      I work inside a sterile laminar flow hood - it filters the air with hepa filters and I clean it before hand with bleach and alcohol

    Is Penecilium not Trichoderma? How to differentiate?

      Penicillium is very similar macroscopically to Trichoderma spp. On a plate Trichoderma will usually spread quickly and typically creates a green, cottony mat, but penicillium usually stays round and uniform because of the nature of it's growth. Further differentiation can be done by using microscopy (penicillium has paint brush shaped conidia "spore-cells" and Trichoderma has more irregular conidia) and growing out on differential/selective medias as well as using specific staining practices. Both organisms are very common and ubiquitous in nature - MUSHLOVE

    What equipment, if any, do you order in bulk? And is there suppliers you recommend or don't?
    I used to do a lot of BRF cubensis (and some straw) years back, so I'm not exactly new to mycology.
    But since im starting up with edibles, I'm going all in on everything I wanted to do before but realistically couldn't because of legality 🙈
    I have a 80sqft room in my basement that I'm making into a lab, if you could do a video about your lab setup (ventilation, materials used, lessons learned etc) I would greatly appreciate it, it might help me avoid a few of the pitfalls while designing it 👍🇳🇴

      Thank you I appreciate the feedback! We have collected all of the equipment slowly over time and have been growing gourmet mushrooms for almost 2 years full-time now - You'd be surprised what you can find on science surplus stores, craigslist and watching patiently for laboratory equipment turnover (I come from a clinical background which is helpful) - We are located in Denver, and are quite lucky to be in such a great market for fresh niche gourmet produce! We are very grateful to get the value for the hard work we put into growing our delicious gourmet mushrooms : ) MUSHLOVE
