I saw the technician in our institute use a syringe to draw out the liquid by unscrewing the screw on the pump.
I am having an issue running Benzoic acid on a Phenomenex Luna C18 column, the peak for the benzoic acid starts eluting fine but then has a really bad tail towards the end affecting my USP tailing, what I don't understand is that my salicylic acid peak is coming out fine as I have both of those components in the same standard solution what can be causing this tailing? I reverse-flushed the column thinking the column was dirty but it didn't help.
Very helpful, is there a way to program a sequence to do all of this automatically and generate a complete report?

Thank for your video. I have problem with my HPLC agilent. Sometimes it can't turn on the light automatically. I turn on the light from the control panel manually. How can repair it to be automatically. My next question is that after 5-6 samples hplc doesn't give right concentration. For example i inject the same vial two times. First time it gives a lot of chroma grams . Second time give the right concentration. What is the problem. I think its problem of the dedector

I'm currently working on a method where a pair of hydrophilic peaks elutes in less than 5 minutes, while another pair elutes about 15 minutes later in isocratic mode. When I use gradient methods (~50 to 70% ACN), the baseline drift increases significantly. What is the best way to run a gradient in these methods? Should I implement a steep gradient change in less than a minute or use a smoother gradient (over a longer period) ?

What about highly acidic or basic samples? Will that affect the C18 ligands of a column. I work with human serum and plasma that are treated with acid, precipitated and the supernate is injected so not the same as the mobile phase.
