at 1:00 you said to draw the blood sample using your mouth or a syringe. Isnt mouth pipetting outdated and really dangerous? we were taught in the lab to never use our mouths to aspirate samples and use an aspiration bulb instead

Use insulin syringe to aspirate.
Why few drop is discarded before charging ??
The pipette is designed in such a way that the liquid in the long tube of the pipette does not take part in the dilution and mixing inside the bulb. So to get the most accurate result, the liquid within this long tube (first few drops) is discarded. Also the liquid in the long tube contains only the diluting fluid. All the cells have been sucked into the bulb. So the diluting fluid in the long tube cannot not take part in the counting and so has to be discarded. Hope that answered ur query.

What 50 consider to cross it with sum of No.of cells in all 4 squares
The formula is : total no of cells× diluting factor(20) /area of squares ( 4×1= 4) × depth (0.1) .... Which on solving becomes no of cells × 50

Extremely sorry for late reply. Dilution factor is simply the number of times the blood sample is diluted , in this case 20 times. So dilution factor is 20. Dilution factor = total volume of sample (blood + diluting fluid) ÷ volume of blood sample taken. So its 100÷0.5
Depth factor is 10, in this case . Depth factor is 10 times of 0.1mm (which is the depth of the space between the coverslip and the counting chamber surface. In order to arrive at the number of cells per cu.mm, we need to make the depth 10times because the area of a counting square is already 1x1mm, so we have to make the volume of a counting square as 1x1x1mm
@@TheSingtangpaScienceGuy nice explanation. Thanks so much. However, I think that "100÷0.5" you wrote as formula for getting dilution factor is for RBC count. In this case, (for WBC count) it is approximately 10÷0.5.
