This screencast demonstrates some of the ways you can change the voicing of a sampleset both by tuning it and alter the volume of individual pipes (this is possible to do on the whole rank/stop as a whole as well).
All changes you make will be saved if you save the organ. The changed settings will follow any selected temperament as you could hear in the screencast.

Will this work on a Yamaha YPG 235 keyboard? it says it has general midi on the kyboard
As long as the keyboard can send MIDI through a USB connection, you should be good to go 😊👍
