This really came in handy for me, I'm having a poster presentation next week at Denver. Important points: 1. Do NOT read your poster 2.Prepare a presentation ~2-3min, and expect questions such as "what do you expect from your research" 3.Handouts are good, you don't have to squeeze everything into the poster, you can have supplemental materials. 4.You can always get back to your viewers. 5. Just admit it if you cannot the question, DO NOT make things up instead. 6. Put the viewers priority.

There's no single answer here, other than assess the situation and respond accordingly. Some people will ask for what they'd like "Please tell me about your work." That's pretty obvious. Some will start looking at your poster quietly with the intent of maybe asking you questions later. You might say "Hi, just let me know if you have any questions." People that approach you rather than approaching your poster - you can tell by what they're looking at - are probably going to say Hello or something like that. You can take that opportunity to introduce yourself and let them know you're available for questions; or you can say "Would you like me to walk you through the work?" I think you get the idea - read the audience, do your best.
