The main person who applies for the visa can only work for the E-2 business, so it's important to structure the business properly from the beginning to include all potential business opportunities. The spouse can work anywhere they would like, including remotely for a foreign business (if permitted by that foreign country). In your scenario it sounds like neither husband nor wife already has a US business up and running.
