Polytene #Chromosome from Salivary glands of Chironomus larvae :
Materials required:
i) 4th instar Drosophila larvae
ii) Two pointed needles
iii) Microscope slide and coverslip
iv) Dissecting microscope
v) Aceto-alcohol (1:1) fixative
vi) Aceto-orcein stain
vii) 45% acetic acid
viii) Compound microscope
ix) Large watch glass or petri dish
x) Blotting papers
xi) 1N HCl
Procedure:
A) Dissection of Chironomus larvae:
1) Dissect the larva with the help of pointed needles as shown in the video.
2) Hold the posterior end of larva with one needle and pull the anterior part with the second needle.
3) Locate the salivary glands and remove other debris.
B) Fixation and staining of Salivary glands of Chironomus larva:
1) Fix the salivary glands with aceto-alcohol fixative for 5 minutes.
2) Remove the excess fixative with blotting paper.
3) Add two drops of 1N HCl over the salivary glands of Chironomus larva to hydrolyze them.
4) Incubate the slide for two minutes at 57 degree Celsius.
5) Remove the excess HCl with blotting paper.
3) Add two to three drops of Aceto-orcein stain over the salivary glands of Chironomus larvae.
4) Cover the slide with a petri dish or large watch glass and wait for 10 minutes.
5) After 10 min. takeout excess stain by washing the slide with 45% acetic acid.
6) Remove the excess stain and acetic acid with blotting paper.
7) Add one drop of Aceto-orcein stain over the salivary glands of Chironomus larvae.
8) Cover it with a coverslip.
C) Squash preparation of salivary glands of Chironomus larvae:
1) To squash, the salivary glands under coverslip place the slide between the fold of blotting papers and apply pressure with the thumb over the coverslip and roll it.
2) seal the edges of coverslip with nail polish or DPX.
D) Observation of squashed salivary glands of Chironomus larvae under a microscope for polytene chromosome.

E.G.Balbiani (1881) first reported the Polytene chromosomes.
Balbiani rings are exceptionally large puffs on the polytene chromosomes in the dipteran Chironomus salivary glands.
I also done it in my lab..and it was very difficult for me to separate the salivary gland of larva..Your work and handling everything is in good and clean way..Thanku for sharing

It was very difficult for me to remove the salivary gland from Chironomous larvae 😓😓😓Sir suggest me some tricks 😓

Believe me, it is very easy to remove salivary glands from Chironomous larvae than Drosophila larvae.
Follow these tricks:
1) Observe larvae under dissecting microscope and locate the Head.
2) Use two sharp pointed needle to hold head segment and remaining body.
3) Pull head segment forward with first needle gently. Don't pull second needle with whom you have grabbed the body.
4) You will see along with head segment, gut is coming out. Don't break it by pulling hard and long.
You will see two rings (Salivary glands) attached to the Gut.
Try till you get it. Best luck 😊👍...

1:1 aceto-alcohol means mix equal parts of each liquid... Example- 1ml of acetic acid mix with 1 ml of alcohol...

Because polytene chromosomes are extended and consist of so much DNA, they are easily visible under the light microscope. A useful feature of these chromosomes is that they have a pattern of dark and light bands, like a bar code, which is unique for each chromosome.
