    Hello, thanks for the explanation
    But I have a question, how will the detector differentiate between the reference and the sample beam, if they both merge and enter the detector as one single beam?

      There is a beam chopper (not pictured) - a rotating disc with sections: an opening, a mirror and a black one. When the disc rotates, it intermittently permits the beam onto the sample, reflects the beam onto the blank or completely blocks the beam from the detector. But I never understood how exactly all that worked.

    My doubt is why's it necessary to use two samples simultaneously?? What if a single sample solution is involved??

    Can we use two detectors ,one for sample n another for reference or single detector is enough?

      Hi +arib shaikh. Thanks for watching the video. Hope you have enjoyed watching it. There won't be a problem to use multiple detectors as you suggested. But the reason to use a single detector is to reduce errors arising due to mismatch of the characteristics of detectors. Every detector reacts slightly different to the same input light that falls on it. This can induce errors during final calculations. So it is a better option to use a single detector as it's characteristics will be matched for both the sample and reference signal. One more benefit of using a single detector is that it saves the cost of the equipment and added circuitry. We hope your doubt has been clarified. If still any queries, feel free to comment.     Can we use the "reference" cell with only air inside the cuvette and do a blank with the "sample" cuvette containing our buffer.

      There are losses of incident beam mainly due to absorption of the sample, but also due to the solvent (matrix), but also due to refractions on each interphase: air/cuvette wall/solvent/wall/air. If you keep the blank cuvette empty you have air/wall/air/wall/air infractions. So, no, the blank cuvette should have solvent (and the matrix of the sample).

    why we use to position one for sample and one for reference if we use same position for sample and reference is that ok or not ?

      In double beam spectrophotometer, the same light radiation needs to pass through the sample and the reference simultaneously. This is possible only if both the analyte solutions are kept in the sample holders at the same instance. So, two different positions are used, one for sample and the other one for reference solution. If same position is used for both the sample and reference, it is called single beam spectrophotometer. Hope this clarifies the doubt :)

The working of double beam photofluorometer will be almost similar to it however with the change that the sample and the standard will be placed simultaneously and measurements are taken.
