The differences have to be on a particular criterion variable. For example you can check if Age of the participants differ by gender. In this case this test is appropriate.

Not only this analysis sir even ANOVAs am using it for non-normal data with help of boostrapping will my study be correct with respect and regards
Well if you over 30 respondents in each group, According to Central Limit Theorem (Naval Bajpai) you can assume normality. Just check that skewness and kurtosis is within the range.

No it doesn't. But balance is considered better.
