But mam when we create tetracycline medium and in that if we gro bacteria then because of tetracycline sensitive the recombinant plasmid contain bacteria will die then what is the use of this method because in knowing weather these is recombinant DNA is there or not we are loosing the bacteria which has recombinant DNA and we are getting the bacteria which doesn't have recombinant plasmid so there is no foreign gene in the bacteria which we get after the both destingzing test so we get useless bacteria after all these hard work?

Waisa nahi hota ... choti choti colonies pe test karte hai pure recombinant coloni pe nahi ..... jab hame pata chalta hai ki ye recombinant hai (beacause that colonie died ) then ham original vali colonie ko ok (accept) karlete hai aur further use karte hai .... hope tumhe samjh aa gaya hoga...
