How are RNA transcripts made if the plasmids remain in the cytoplasm?

Great question, thanks for asking. The plasmid contains the information needed for transcription (eukaryotic promoter sequences, etc). To be transiently expressed the DNA can be in the cytoplasm, but will be lost eventually. It is possible to select for stable transfection, in which the DNA has been incorporated into the genome of the host cell.
