What you did with the smoke machine is a very valid form of testing. It's literally called a "smoke test" and is used to detect leaks in air tight systems. It's also where the software industry took the term for a basic test pass to check that everything meets minimum stability requirements
1. VOCs are gaseous and will disperse into the air. You'll need to seal that leak. A bead of caulk around the seams should make it airtight. 2. You can solvent weld acrylic. What solvents you use depends on what results you're going for, I think. 3. Smoke tests are a valid way to see if something is airtight and if the airflow is right. Cool project! Did you do the smoke test while running the fans? Because i wouldn't be so quick to make it completely airtight. Otherwise your creating a vacuum with the vans running and you won't have the airflow to move the voc's. Really the van running should be enough to create a lower air pressure inside, this ensuring the voc's will get stocked out by the van.
You know what, now that you mention it, I'm absolutely certain that one day I'm going to place my cup of tea inside that enclosure and then be mad for hours trying to find it while it's all closed off and printing stuff. So, I should probably add that. XD

The only thing I can really tell you is my personal favorite workflow; however, I can't say with certainty if it's the best or if it's good for everyone. My all-time favorite CAD software (modeling software) is Shapr3D. It works on the iPad, and it's so intuitive that I rarely need to watch tutorials to learn how to do things. Absolutely amazing software! In terms of materials, I've only just started working with resin, so I'm not sure what the best options are. For FDM printing, I prefer PLA. Some people who are much smarter than I am in the comment sections of my videos have also recommended PETG, I believe. Sadly, I mostly focus on build projects on my channel, so I don't have guides or anything like that, but I hope this helped a little. :)
Shapr3d is a pretty solid tool. Pun not intended, but it is parametric solids. Manipulating the view and models on a tablet is incredibly intuitive, even more so than Fusion with a Spacemouse.
