I'm not sure how well tested or consistent it is but right when you start playing the leaf that kills you can be jumped off of and wall kicked, which gives you enough speed to completely skip the "Aunty" boss fight, which can relatively save around 40-50 seconds in a speedrun

After that, is to get through the Supermarket stage where you eventually face your grandma as the boss fight. First you have to avoid talking by moving 3 steps back so she moves closer.
