We are applying compression in the vertical direction but the cylinder will undergo tension along the length.

Yes, unless your aggregate size is larger than allowed for a 4x8.

I have done this test after treating the concrete with different conditions to see which treatment improves the concrete.

According to ASTM you are supposed to average two measurements of length.

The test is an indirect way of determining the tensile strength of concrete since direct methods are somewhat difficult.
