No only chosen subjects

No. There is no 'compulsion' to take any other subjects. However, you may appear in as many subjects as you wish.

You need to apply at any college/school offering A-Levels. They usually have a certain merit for this.

I really don't know much about colleges in Karachi, but accepting a Matric student for A-Levels depends entirely on the college's policies.

Most of colleges do accept and also offer good scholarships
