here we find work due to externally applied force...angle between this force and displacement is 0 degree... actually -ive sign arises due to opposite directions of electric force and externally applied force but these two forces are equal in magnitude because we have to maintain electrostatic equilibrium.

Because charge is moving against electric field which is opposite and opposite angels is 180 which gives -1 meaning negative sign is for moving against the electric field

Appreciation for explaning the gradient.Sir most of the teacher leave the basic concept such as;
1:what is point charge?
2:what is rate? Etc
When distance btw the charges is greater then their size this is called point charge or localized charge.
