A tad confused on inputting the data onto prism, i can see a column format in your video, where do you place, say, 3 data replicates for each condition?
Hi :) to enter replicates, in the type of graphpad spreadsheet, choose "grouped" and on the options select "enter x replicate values in side-by-side subcolumns". Let me know if this helps.

you didn't actually show how to perform the Kruskal-Wallis analysis
Please check the video from minute 2. It's the "non-parametric" option in the "assume gaussian distribution" menu.

Yes I believe you have to represent it by mean. I'm sorry it took so long to answer
