ah, this makes it look so easy. My cover slips never come off the well this gently :')
And sometimes they come off so abruptly they do several flips and I lose track of which surface the cells were on..

One of my mentors suggested a really handy trick for doing this. You take a needle from the syringe. You bend its free end inward so as to make an L with a tiny foot. You use this L to lift the coverslip up and then carry on with your usual business using the tweezers.

@@vishweshrai Oh yeah, I actually use needles to do the lifting too. And was taught the same thing. I never could figure out how to bend a needle though, so just kept on using it in straight form.

    1) are the cells on the coverslips stained ?
    2) what mountant did you use ?

This is handy to show people, but every time I have to explain they use way too much mounting media in the video

Can someone tell me a protocol for mounting live cells grown on a coverslip onto a slide?
Also, if I am using a live fluorescent probe and cannot fix my cells, how do I proceed for counterstaining with an antibody?
    Thnx in advance

How to sterilizer the cover slip? Can i just dip it in to alcohol? 🥶 thanks

Yes, you can dip the clean coverslips a couple times into methanol or isopropanol. Then, let it dry in a petri dish inside the hood. Optionally, you can go a further step by passing them quickly through a flame after the methanol dip. Hope this answer was helpful.

what solution do you use for incubating the coverslips before mounting???? thanks for the video btw

    • @thermofisher

Cover slips do not need to be incubated in any type of solution prior to mounting. They can be taken straight out of the box to use. Please let us know if you have any additional questions and we’d be happy to assist.
