Sir I have 1 question regarding non competitive and uncompetitive inhibition
Why km value is not changed in non competitive inhibition because when inhibitor binds with ENZYME or ES complex ( binding site for I and S is Different) but inhibitor will not decrease the affinity for substrate, because it cause some conformational changes at catalytic site ???
In noncompetitive inhibition
Inhibitor does not affect the binding of substrate to the enzyme. It's efficiency to convert the substrate into the product is decreased.

In vasudevan, non competitive inhibition is given in irreversible and also written that inhibitor combines with enzymes by forming a covalent bond. Sir please clarify it.

Content is taken from Tietz textbook of Biochemistry. Non-competitive inhibition is sometime irreversible. In some books Competitive and Non-competitive mentioned while in some non-competitive is mentioned under reversible inhibition.
Vasudevan is good book. Just follow any one book for this topic.
That's a flaw of research. Scientists are divided in two groups regarding this
