Why cathode and anode is required here???what does it do in separating the particles
It measures the flow of currents towards the orifice.In simple words wwithout those you cannot calculate the change of resistance on the orifice and therefore the amount of cells going through the orifice

Size of orifice???
depends on size of particle being measured. It's diameter must be just greater than that of the particle but less than twice it's size to ensure only one enters at a time, I think. I'm not sure about this but this is how I understood it.
