    Iodine is used as a stain to help you better see the cells. In this video, we are viewing elodea leaf, which contains the photosynthetic pigment chlorophyll and appears green. Since the cells already have green color, there is no need to stain.

      +Megan Beckford One of the most common problems when first focusing on s specimen is not bringing the stage up high enough towards the objective lenses. When on 100xTM (yellow banded 10x lens), use the coarse focus to bring the stage all the way up, then lower it a bit. Now you will be in the "neighborhood" of a clear image. Hope this helps.

    Why do we need to put water in the leaf instead of putting iodine solution ?

      +ItaaMashita The leaf is already green from the chlorophyll, allowing you to see the cells. No stain needed :)
