How about a threat who challenges Po with love. Love can be your greatest strength but also your weakness. And the fact that Po wasn‘t in love would be nice to see. He always had a crush on Tigress and the chrush could turn into real feelings for her. He could be challenged in romance. Either something happens to her during a fight with a villain who was in love with her in the past, when she was a teenager or her past (there must be a reason why her parents left her in the orphanage) will come haunt her back and Po will become emocionally confused.
I have a theory about the last two films in the franchise: Kung Fu Panda 5: Mantis, Viper, Crane, Tigress and Monkey will train their successors as next Furious Five, and Zhen officially become the next Dragon Warrior thanks to Po Kung Fu Panda 6: Shifu will pass away and Po will become the new Senior Master of the Jade Palace to take care of the next Generation of Kung Fu
