    Note that PPQ is just an example molecule and likely has no effects through oral ingestion. It would largely be a sedative which is unlikely to give a pleasant effect like methaqualone, its activation of receptors is different than that of methaqualone to the point where it is unlikely to produce the right dopamine receptor activation.

      We've known how to make Quaaludes for ages.
      There's a bunch of reasons no clandestine chemists produce it outside of South Africa.
      1. The dose size is pretty big so you need large volumes of heavily watched precursors to make it.
      2. The market is surprisingly limited, sure if you've seen the wolf of wall street you might try it but even then they'd probably say its fake considering how rare it is.
      3. There's plenty of far more profitable and popular chemicals to make, why make Quaaludes when you can make MDMA?
      The only reason its even produced in South Africa is because the Triad smuggles some weird fish from there that's a delicacy in China and in return they smuggle back the precursors for making bathtub Quaaludes.

    We had this kid at my drug rehab who knew how to make these and he called them "lemon drops." A lot of drug tests were simple 6 panels. So they never showed up. He ended up overdosing on benzos and pain killers. But he was a smart dude

    If the estimate of an active dose of this is 0.5mg, this synthesis at 13.5g is 27,000 doses! More importantly, it's not hard to absorb such small quantities through the skin or mucous membranes, so PPE would be absolutely essential in replicating this.

      I think skin absorption is generally overstated. The skin acts as a pretty good barrier especially to larger compounds. You would have to apply something like dmso to increase permeability to actually be worried about accidental ingestion. Still PPE is always a good safety precaution.

    Have you seen any literature on the reaction straight to the benzoxazinone with anthranilic acid, benzoyl chloride, and pyridine? I have tried this reaction but was not able to get a product with the correct melting point. Eliminates the need for DCM and triethylamine based on multiple papers I’ve seen, but I haven’t gotten it to work after a couple goes. Great work on this stuff man!

      Could theoretically work if you added at least 2 eq of benzoyl chloride, which would acylate both the amine and create the mixed anhydride with anthranillic acid. The mixed anhydride should be activated enough such that the amide carbonyl can attack and form the benzoxazinone. I'm not exactly sure the stoichiometry Chemiolis used in the first step to get the single acylation product but the excess acetic anhydride in the second step essentially activates the acid and dehydrates the rxn driving to completion. Again on paper this looks like it would work, would have to try in lab.

      Have a go using isatoic anhydride.
      I had the same issue as you, but had success with that.
      The route is toluene to nitro toluene, to o-toluidine (separation is less painful than anticipated), then you can go to N-Acetyl-anthranillic acid or isotoic anhydride.
      It’s weird seeing Americans give each other advice on avoiding hard-to-get stuff.
      You guys can get concentrated HCl OTC, but can’t get pyridine or piperidine or cyclohexanone.
      In the UK I can get everything except strong acids OTC, and to get strong acids all I need to do is create a company (that has a legitimate need for the chemicals - research ticks all the boxes).
      I could get a canister of HBr gas as a 17 year old with an amateur lab, and when the police raided me they were only allowed to confiscate the 25% H2SO4 lol. They had to give it back, don’t worry.
      I was able to get several litres of oxalyl chloride and PCl5.
      I remember going on sciencemadmess and asking for input on a paper claiming “metal acid anhydrides” (eg Fe2O3 - I know, weird) could be converted to the Lewis acid trichloride with Oxalyl chloride.
      And they were furious - they were trying to make oxalyl chloride with thionyl chloride or AlCl3 and here I was with litres of the stuff lol.

      We can't get cyclohexanone, pyridine, or piperidine since they're such common base structures of recreational drugs. Piperidine alone gets you within a couple of steps of making phenethylamines, and we can't have that! 😮‍💨

Almost every old women their has been on Valium since the early 1970's and all are extremely healthy and mentally vibrant!!! They are all sharp and sassy and do not have any cognitive problems. With likely similar levels of recreational use to less euphoric benzos or even possibly antihistamine sedatives

      Ziconotide is a pain medication that doesn’t cause physical dependency, but it has terrible side effects and can only be administered via an intrathecal pump. It’s a selective N-type voltage gated sodium channel blocker. Personally, I’m just fine with opioids being the mainstay of chronic pain treatment, humans have been using opiates for thousands of years. I think the reason why we’re seeing such an increase in opioid deaths is because doctors stopped prescribing them and kicked so many people off of their prescriptions.

      That, and the increasing prominence of fentanyl, and its ~50 analogs. Nitrazenes are also becoming more common because they are harder to detect, cheap, and very simple to synthesize (basically the same reasons fentanyl analogs became very common). These nitrazenes are benzimidazole derivatives that have even greater affinity for the δ-opioid and μ-opioid receptors than fentanyl (they were discovered in the 1950’s, but the researchers decided they were far to potent to be safely used in medicine. Thus, they were never approved for use). The consumers of these types of chemicals expect diacetylmorphine, oxycodone, etc., but they instead unknowingly end up with chemicals that are hundreds to thousands of times more potent. The more potent these chemicals are the less effective naloxone is at reversing the opioid toxidrome as well requiring larger quantities, and more frequent repeat administrations of naloxone. Currently there isn’t a test kit for nitrazenes. Fatal ODs have about doubled in the last decade because of these factors.
      With all of these factors combined it’s really not surprising that we’ve been having this serious problem. This will continue to be an endless cycle until serious policy changes are made, and new strategies/protocols are adopted. Helping patients ween off these medications when it’s no longer needed, and making naloxone readily available everywhere would definitely be a great start though.

It Never made sense to me how they said they wanted to treat the opiod problem yet they completely stopped giving pain treatment and didn't think that would back fire and cause a ton of people to seek other illegal options through out the black market.. I do think it is possible to have therapeutic pain management treatment but apparently our society doesn't believe that exists..

We could make workplaces safer to prevent more injuries that cause chronic pain conditions. We could provide better psychological and medical treatment for dependency and create a culture more receptive to those who are dependent and wish to end their dependency without labeling legitimate pain patients addicts or using accusatory language such as "drug seeker" based on limited interactions with patients during single encounters. We can make methadone treatment more widely and easily available to those seeking treatment.

    Chemolios is one of the best chem TH-camrs by far. One thing I always wonder when I see his videos is how the hell does he aquire some of the chems? I know for sure some of the things he has used isn't commercially available to the home hobbyist. I'd like to have a combo with dude. I have some ideas I'd like to see one of the chemtubers tackle. Hell Ive though about shooting a vid myself but I think someone like chemolios would do it much more justice.

    Other analogues like cloroqualone, etaqualone and diproqualone were developed in france from 1950 to 1966. Used as antitussives, muscle relaxants for neuralgia, mild anxiolytics and moderate potency hypnotics. But was pulled off market by 1970 due to reports of abuse and toxicity. Other analogues were manufactured in clandestine labs jn Germany and Poland for use as street drugs. Some analogues are still prevalent in Underground Rave clubs in Russia as cheap dissociative drugs, cheaper than ketamine or etomidate or xylazine. The downside of all these analogues is the eventual renal disorders and the occasional cerebral inflammation and death, if you od'd.

      @@_Y.J yeah I’ve done em all and xans made me black out the most but still fully functional and running only on my subconscious lol not cool at all I don’t recommend to anyone reading this. I liked Valium because it had a unique feeling to it that was like a warm blanket filled with pure love and also gave me the munchies worse than weed does. But Librium is king of them all and only given to severe alcoholics because it lasts pretty much 24 hours damn near and comes on slow but has a strong peak that lasts without making you black out like xans but just as strong only mellow come up so you get used to it without it hitting you all at once. Temazepam aka Restoril is a combo effect and very euphoric as well. Feels a lot like Ambien but with a Valium like feeling and also lasts longer than Xans do and also stronger imo. I never tried “Research” Benzos but I heard they were legal to buy for years online until people started yappin their mouths just like with the pain meds and they made it illegal to get online from that gray area loophole. I also tried pure phenibut from “Nootropics depot.com” before they banned it and that shit is a hell of a lot more dangerous and worse withdrawals than any benzo or alcohol I ever done lol phenibut loses it’s magic quickly and the withdrawals are far beyond anything I could put in words. Vivid hallucinations and extreme auditory hallucinations as well hearing voices and people not there while fearful of closing your eyes because as soon as you do it’s full on cartoon like *scary* hallucinations so vivid you can’t tell it from this reality. The extreme sweating and convulsions, restless arms and legs with mild seizures just aren’t worth taking pure phenibut for more than 3 days straight. Scary shit and the panic and fear are on a level you don’t wanna reach lol who ever reads this “don’t take phenibut” most is fake now days anyways or cut with shit that will mess you up.

      Definitely nostalgia plays a huge part in how they remember qualudes.. I’m 34 so I definitely wasn’t around but I’ve been a Xanax (and kpin) connoisseur since o was 18 very experienced with benzos and I actually got my hands on a legit qualude I know someone who knew someone who had some back in the day and stashed them before he went to prison got out and they were still were he had them gave some to my boy who I got it from ,, literally it felt very similar to alprazolam same thing pretty much

      @@amritlohia8240 that could be about right the dude was older probably if he’s not dead 60s so if he got them towards the end let’s say 1980 stashed them and got locked up right after did let’s say 20-25 I do t remember but I know he was in for awhile it wasn’t some 3 4 5 6 7 years I know it was double digit n I wanna say in the 20s mark somewhere ,, n got out early 200s let’s say 2002 went to his stash spot got em and I know he had a lot because he was a dealer back then my friend said , then sat on them for roughly 10 15 more years using them special occasions and only giving limited amounts to close friends and then my boy got em he had only got like 4 or 5 of them and gave me the one somewhere round 2010 ,,,, that’s probably what happened ,,,, or it all could be completely made up story some tweaker told my friend actually gave him some pressed Xanax or some designer benzo in form of a lude

    8:19 - It wouldn't surprise me if the acetic anhydride acetylates the carboxylic acid first to form the mixed anhydride, which is then more susceptible to the intramolecular cyclisation. Acetic anhydride seems like an odd choice of solvent otherwise.
    I wonder if it's possible to use excess benzoyl chloride to acetylate the nitrogen, form the mixed anhydride and then cyclise all in one step...

      Neat! Yes you can do that, I have done that same cyclisation before for a 3H quinazoline, you do need a drying agent since it generates water - the best out of 6 tested was PCl3 (since it dissolves better in MePh than PCl5)

    You should do an an anthranilic acid synthesis video from methyl anthranilate which would be a simple demethylation, idk the exact chemistry involved but you probably do and the precursor is widely available as artificial grape flavoring

      The reaction is base hydrolysis, simply stir and reflux 151 grams of methyl anthranillate in a NaOH aq solution made from 40 g of sodium hydroxide and ~ 300 mL of water. This makes sodium anthranillate which dissolves in the water along with methanol. Then adding 120 grams of 32% Muriatic acid precipitates the anthranillic acid which is insoluble in water.

    Why not just stay with the original Quaalude? Hugh Hefner called them "thigh openers".
    Take of {acetyl-anthranilic acid}, add {ortho-toluidine}, mix well with polyphosphoric acid and heat to about 250.
    Quench into water, basify, extract with organic solvent, wash a few times.
    Add an equivalent of HCL and crystallize.

    So the step where you ad acetic anhydride and then the next step where you add aniline don't need a separation work up. You can just add the aniline to the mixture with some extra amount of acetic acid, since acetic acid is the solvent and separation and purification seems like it just reduces your yield.
    Also, the final purification steps should just be toluene. you don't need the dcm stuff.

    Honest questions about the synthesis.
    1) why wasn't the acid chloride added dropwise to the mixture of anthranilic acid and triethylamine? Surely the mono acylated substrate is sterically hindered but I'm expecting double acylation on the amine
    2) Was water removal using acetic anhydride described? Also here again I expect acylation of the amine and potentially forming mixed anhydride with the free carboxylic acid which can cause your substrate to form inter molecular acylation reactions. Edit: your over 100% yield is coming from that.
    3) why do you never show TLC? It's so fast and easy
    Not trying to bash, just curiousity.

    I think the mechanism can be another: anthranilic acid is a zwitterion, TEA deprotonates that which amino group can then attack benzoyl chloride. I also think about TEA attacking benzoyl chloride making a quaternary ammonium which is a great leaving group displaced by amino of anthranilic acid. I still wonder where do you get all these restricted chemicals.
    Can you tell me something about fluorescence of PPQ ?

      Id guess it's because you cant immediately die from benzos alone. I believe the long term effects and withdrawal of Xanax to be among the worst of any drug ive ever encountered.. and I've done nitizenes and iso which is worse than fentanyl. I would rather smoke fentanyl for a year and cold turkey than i would take Xanax for 3 months and cold turkey.
