Fun Fact: This fight with D'arby inspired the creation of Yu-Gi-Oh! In the beginning of the original manga they played all different kinds of games. Only after the chapter with the card game (created as a parody of the oldest trading card game, Magic: The Gathering) that featured Kiba became so popular with people, who also wanted to play it IRL, did it become an actual card game with the manga switching to focus on it.

So Jotaro's bluff is perfect because you have to remember the type of guy Dio is. Obviously if D'Arby calls and loses Dio will kill him for releasing that info. But at the same time if he calls and wins keep Dio's personality in mind. "You wagered WHAT on that game of poker?" Jotaro instantly made it into a lose lose situation for D'Arby, he had no option but to fold.
