    Just had my practical this past week. I did well, but I wish I watched this tutorial to give me an extra boost of confidence. I will use this for my quiz on Monday lol. Either way great tutorial and explanation. Thanks!

    Why not talk about adjusting the iris diaphragm for improved contrast and resolution. To remove air bubbles in the oil, rock the 100X objective back and forth through the oil.

    You should never rub the lens that has been immersed in oil. The trick is to dab it with a special paper until its oil free.

    wiping slide with paper towel will also remove the smear, so bad idea to do .

    do you do place the oil directly over the fixed bacteria or do you use a glass cover plate over the bacterial?

      in this course that i am taking right now we put the oil right over the fixed bacteria but in a previous course the instructor told us to put the oil with lenses paper on the lenses it self, personally i find putting oil directly on the bacteria works better tho it can get messy if you move the stage positions.

      prettymuchevann just use your shirt or whatever is around.
      It's not your microscope right?

    Very well done; taking off the immersion oil with a real lens cleaning paper, but when you are not sure that your 100x is clean ALWAYS use a good quality lens cleaner (like ROR) to really clean your frontlens! NEVER clean it dry! RoR is one of the rare cleaners that really resolve parts of immersion oil, instead of dilliputing it!

    Ma'am I have a question. Can we use any sort of oil or is there a specific oil only meant for this job?

      You should use an immersion oil that has a refractive index equal to the refractive index of the lens.

      mScope ML-A-A Microscope Immersion Oil and you need kimwipes lens cloths and Hexane to clean the lens after you use it.

    can I use mineral oil or baby oil for the oil immersion obj?

      We actually did a research project on that and we used 3 oils as substitute and compared them if which one is the best to use as an alternative for immersion oil. It was virgin coconut oil, baby oil and castor oil and according to what we actually performed, the castor oil gives the best results. It is so clear and almost as the same as the immersion oil.

      NO..Use only immersion oil. It has refractive index of 1.515. Same as glass cover slip and slide !

    The 100x oil immersion objective will work without oil just fine, but the image will have reduced contrast and resolution.
    The oil isn't neccessary and nothing bad will happen if you don't use it. -_-

    1:18 did you use prepared slides to demo the oil immersion? Prepared slides are only designed for dry objective. 😂😂😂😂
