It's a cool philosophical and sociological examination of just how obsessed our society is with knowing what gender category to place people, even in a situation where it doesn't actually matter, like a co-worker's birthday party during office hours.
It's not sociology, it's biology, and it always matters, even at a birthday party. It's 2 million years of evolution that has hard wired us to judge each other and treat each other based on knowing each others sex. That is why our sex is usually so easy to discern, because it gives us an evolutionary advantage.

Thanks to the movie, we know that Francis is not Pat's mother. This is the father. Not just because this is a different actress. But in the movie, when the doctor is about to say Pat's gender, he is cut off by a nurse who needs help. The way Francis describes being told the gender is different. To be honest, I think you make too many assumptions about Conservatives.
You will find they are a much more diverse group than you have stated - they're not all racist, homophobic bigots who cling to their Bibles and never saw a war they didn't love.
In fact, they are more likely to place greater value on the freedom of an individual to express their individualism than most (collectivist minded) Progressives actually do.

It's still funny
Thanks to the movie, we know that Francis is not Pat's mother. This is the father. Not just because this is a different actress. But in the movie, when the doctor is about to say Pat's gender, he is cut off by a nurse who needs help. The way Francis describes being told the gender is different. I know saying this ruins the joke of the video, but hey. This makes one mystery solved on who Francis/Frances was.
My favorite Pat Skit is when Pat.goes to the Gym to see if Pat would be interested in a membership. Pat has the Gym Manager so frustrated when it's over. I laugh so hard every time I see it
