The dependent variable is the presence or absence of starch. There isn’t an independent variable in this demo, but you could easily construct one by changing a factor, eg. covering a part of the leaf, removing CO2 etc.

Iodine forms a blue/black complex with starch, which is left in the leaf, not chlorophyll
If we remove chlorophyll why is there still starch present?

Why was alcohol used to treat the leaf?
This removes the chlorophyll, turning the leaf white so that the a colour change is easier to see during the iodine test.
