Masson's Trichrome Staining method is used for differential staining of collagen fibers and smooth muscular fibers in tissues such as skin, heart, etc. on formalin-fixed, paraffin-embedded sections, and may be used for frozen sections as well. As a result nuclei will be stained in black color, cytoplasm, keratin, muscles - red, collagen - blue, erythrocytes - yellow.
Masson's Trichrome Staining Protocol:
1. Weigerts hematoxylin - 10 min.
2. Dry slides without washing.
3. Alcoholic solution of picric acid - 4 min.
4. Wash the slides quickly in distilled water (3 - 4 sec).
5. Acid crimson fuchsin solution - 4 minutes.
6. Wash the slides in distilled water.
7. Phosphomolybdic acid - 10 min.
8. Dry slides without washing.
9. Massons anyline blue - 5 min.
10. Wash slides in distilled water.
11. Rehydrate, clear and coverslip.

Can we use tap water instead of distilled water? Is there any critical issue by not using distilled water?

How can i get it? Where
how long it takes to do that and a waste of reagents
The staining protocol takes about 30 minutes. In this video we used a kit for 100 tests, also there are kits for bigger quantities of tests and they are usually used for automatic stainers.
Do we have to dewax first?
Please what stain can I use to demonstrate mitochondria?
For mitochondrial muscles patology u can use gomori tricrome, If I'm not wrong
