Composition and prepration of 50x TAE Buffer stock (Tris acetate EDTA)

  • TAE buffer is a buffer solution containing a mixture of Tris base, acetic acid and EDTA. In molecular biology it is used in agarose electrophoresis typically for the seperation of nucleic acids such as DNA and RNA. It is made up acetate buffer, usually at pH 8.3, and EDTA, which sequesters divalent cations. Here we have made 50x TAE buffer and mentioned a step wise protocol for prepration of 1 litre stock, for 2 litre you have to double the measurments and quantity of chemicls used.     The method is from sambrook. I am keen to know about how you measured EDTA as 0.5M and maintained pH of 8.0

    What is X in 50X??
    I mean what does X denote here...plzz tell i have my exams today

      X basically denotes the concentration of the solution, like (50x) means 50 times!
