Chlorophyll isn't soluble in water. It is soluble in alcohol, when boiled. We want to remove the chlorophyll so as to see the iodine marks clearly at the end of the experiment. Hope you understood.
By the process of photosynthesis the starch was formed , so we can say that if the part contain starch it was gone through the process of photosynthesis.

The part of the leaf is covered With black paper in order not to let it be exposed to the Sunlight.....Thus We can conclude sunlight is needed for photoshynthesis as in the last we have observed that only the part of the plant that is exposed to sunlight turns blue-black in colour(The part that is not exopsed do not change its colour because photoshynthesis do not take place)........
Due to keeping the plant in dark room starch consumed by plant. If we test the leaf with iodine total leaf does not turn into blue blak colour due to the absence of starch in leaf. So we must keep the plant under sunlight for production of starch in leaf. After that only result come perfect.
